Prepared from Elastin EPC No. E60 by proprietary modifications of the method of Mecham and Lange (5).
Generally, bovine neck ligament elastin is extracted two times for 60 minutes at 37º C with 1 M HaOH om 80% ethanol.  The second extraction supernatant is neutralized with HC103,  clarified by centrifugation, dialyzed and lophilized.  ETNA-elastin is a salt-free, water soluble powder.

AB415, Bovine Aorta Strips are designed to be shipped at amibient temperatures, but require special conditions and storage.  Immediately upon arrival, store at the temperature indicated on the product label.

This is the starting elastin used for the preparation of all chromophore-labeled and fluorophore-labeled substrates described herein.

Elastin E60 is purified from bovine neck ligament (EPC No. B214) by the neutral extraction method of partrige et. al. (1). The method yields a product that is free from fat, muscle, collagen and connective tissue.
Assays of nitrogen, hodroxyproline and amino acids are used to monitor the purity during and after preparation. The final product is characterized by consistent chemical properties and optimum reactivity with elastase. It contains less than 0.01% insoluble residue after elastolysis.

The rat lung elastin is purified by the method of Starcher et al. (3) in which contaminating proteins are removed by mild extraction conditions. This method minimizes peptide bond cleavage.  This elastin is insoluble.

Coacervation is repeated two times more in order to improve the purity of the alpha elastin fractions.
Alpha elastin has a molecular weight of 10-60 KD and runs as a smear on PAGE.

In the method, the coacervated alpha elastin is separated from the soluble beta elastin at pH 4.7 and 50° C. Coacervation is repeated two times more in order to improve the purity of the beta fraction. Beta Elastin does not coacervate.
Beta Elastin has a molecular weight range of 3-10 KD.
