所需材料
pluriPlix® (50-01010-80)
10x Wash Buffer 500 ml (60-00080-11)
Density gradient medium*
Label-标记
1.重悬pluriSpin®,旋涡20秒;
2.向全血中加入pluriSpin®悬浮液,浓度为50 ul/ml全血,旋涡2秒。
Incubate-孵育
3.室温下,在pluriPlix®或滚动搅拌机上孵育15分钟,15rpm/min;
4.用等量的洗涤液稀释样品,轻轻混合。
Layer over density gradient medium-平铺密度分离液
将稀释后的样品平铺在密度梯度介质上
注意:注意尽量减少密度梯度介质和样本,体积建议见下表。
Spin-离心
6.室温下800×g离心15分钟。
Collect-收集
7.小心地从密度梯度分离液层和血浆层中间移走富集的细胞；
8.将富集的细胞加到新的离心管中，涡旋5秒，避免聚集。
Wash-洗涤
9.在富集细胞的离心管中加入洗涤液；
10.4℃,300g离心10min;
11.去掉上清，并重复洗涤一次；
12.重悬细胞沉淀。
